How do you ProLong gold Antifade reagent?
Product Usage Information Following immunostaining, cover samples with ProLong® Gold Antifade Reagent and coverslip. To prevent fading, allow slides to cure in the dark, at room temperature for 24 hours, then seal the coverslips with clear lacquer, such as nail polish. Proceed with imaging.
Does ProLong gold have glycerol?
When comparing this product to the competition’s mounting media, keep in mind that ProLong® Gold is a glycerol-based hard mount media. This means that it cures over 24 hours to a solid state so coverslips do not need to be sealed to the slide with products like nail polish or beeswax.
How do you ProLong the diamond Antifade Mountant?
Just apply a drop to the sample, add a coverslip, cure, and image. ProLong Diamond mountant is a curing mountant that forms a near-perfect optical path (1.47 RI) and enables longer-term storage of your samples, while protecting them from photobleaching.
How long does ProLong gold take to cure?
24 hours
For samples mounted using the ProLong™ Gold or Diamond Antifade Mountant, allow the preparation to cure on a flat surface in the dark. Curing time may vary from a couple of hours to overnight, depending on the thickness of the sample and the relative humidity of the surrounding air. We recommend 24 hours curing time.
Does ProLong gold Harden?
Our ProLong antifade reagents dispense as a liquid that will solidify upon the evaporation of water. SlowFade antifade reagents remain liquid. If you are going to image right away and then dispose of your sample, you do not need a mountant that hardens, such as the SlowFade reagents.
What is vectashield?
Description. VECTASHIELD Antifade Mounting Medium is a unique, stable formula for preserving fluorescence. This medium inhibits rapid photobleaching of fluorescent proteins and fluorescent dyes.
What is mowiol?
Mowiol® is a water soluble hydrocolloid mucoadhesive based on poly (vinyl alcohol). Mowiol® at pH 8.5 has a couple of advantages over other mounting media: its a liquid when the sample is mounted and solidifies after several hours, forming a permanent bond between the coverslip and slide.
How long does vectashield last?
two months
Most recent answer We use the Alexa Fluor, Vectashield + DAPI, 4 ° C in the fridge for two months of storage in a humid chamber with PBS.
What is DPX chemical?
DPX is a synthetic non-aqueous mounting medium for microscopy. A traditional resin-based slide mountant with xylene solvent. Of medium viscosity and is touch dry in approximately 15 minutes.
How do you make mowiol?
MOWIOL Protocol Heat 12ml of 0.2M Tris, pH 8.5 to 50°C in a water bath. Add 12ml prewarmed 0.2M Tris (pH 8.5) and keep at 50°C for 10min. with occasional mixing. (Mixture can be kept at 50°C for 1hr with agitation every 10min.
How do you use Entellan?
Use: Cover the object with drops of Entellan® so that the space between the cover glass and the slide is completely filled. The cover glass must be sealed right up to each edge. Excess embedding agent can be removed with a cloth moistened with xylene.
How long does immunostaining last?
It depends on the fixation and mounting protocol. I usually fix animal cells or sections with 4% PFA in PBS or methanol. For embedding I normally use an embedding medium containing glycerol and N-propyl-gallate (anti-fading reagent). With this protocol the fluorescence will stay relatively constant for several months.
Does vectashield need to be refrigerated?
The original VECTASHIELD Mounting Medium does not solidify, but remains a liquid on the slide and can be stored without sealing. If desired, coverslips can be sealed around the perimeter with nail polish or a plastic sealant. Mounted slides should be stored at 4 °C, protected from light.
